Scope of Work
The NIH seeks a confocal microscope system for advanced biological research, specifically for imaging neuronal dendrites approximately 1 µm in diameter with dim fluorescent signals, requiring high-speed imaging and a high signal-to-noise ratio. Key requirements include:
- Configuration: Upright microscope, compatible with high-numerical-aperture water-dipping objectives and simultaneous patch-clamp electrophysiology.
- Detector Technology: 25-element SPPC array detector with single-photon sensitivity and ultra-low noise, offering improved SNR over conventional detectors.
- Imaging Capabilities: Supports live imaging of ex vivo cochlear preparations, time-lapse, calcium, neurotransmitter, and voltage imaging. Must enable low-light imaging, reduce photobleaching and phototoxicity.
- Performance: Approximately 100 nm lateral and 300 nm axial super-resolution, compatible with AX R resonant scanner, pixel dwell time as short as 200 ns, up to 720 fps acquisition, and FN25 optical system.
- Software & Integration: Compatible with Nikon NIS-Elements software, existing Nikon imaging workflows, and capable of supporting NIH and extramural collaborations.
