Scope of Work
The USAFA Department of Chemistry requires a replacement for a 20-year-old fluorimeter whose excitation source is failing and for which replacement parts are unavailable. A modern replacement is critical for upper-division course instruction impacting 60-80 cadets annually. Key requirements (salient characteristics) for the new system include:
- Multiple Data Collection Modes: Must support fluorescence, phosphorescence, chemiluminescence, and bioluminescence.
- Excitation Source: Xenon flash lamp-based system, minimizing photo-bleaching, with a high-frequency flash rate and room-light immunity.
- Optical Beam Geometry: Horizontal configuration to maximize collection efficiency for low sample volumes.
- Sensitivity & Detection: High-sensitivity photomultiplier tube for enhanced sensitivity above 700 nm, capable of measuring picomolar amounts of fluorescein, with a guaranteed Signal-to-Noise Ratio (SNR) of >500:1 for the Raman Band of Water.
- Sample Handling & Temperature Control: Automatic measurement of four or more cells, includes solid sample holder accessory, and a water-thermostatted single-cell holder with a compatible water pump. Integrated Peltier thermostatting system with ±0.3 °C accuracy.
- Data Collection: Microsecond time intervals for phosphorescence; capability for fluorescence scans at different temperature intervals.
- Sampling Versatility: Capable of reading microplates for high-throughput analysis and solid samples.
- System Configuration: Includes a desktop PC with compatible software for instrument interface and control.
